It was one of those days when I doubted my worth while doing undergraduate level kind of menial labor. There is no thought process involved or problem solving or any high level of even general thinking involved.
76 strains of bacteria are going to be shipped off for sequencing in the next few weeks which is my 'project' while I wait for my other ones to get back on track.
69 samples of culture was aliquoted from the chamber. A good two hours of work. Labeling small tubes is a bitch by itself that takes half hour.
69 samples for crude DNA extraction. Another two hours.
Another two hours of preparing 68 PCR samples. I lost sample 220 somewhere in my melting ice bucket or overcrowded water bath. Knowing me, I'd probably find the pocket of my jacket tomorrow.
3 hours of waiting for the PCR to end. Which was okay because Potbelly was quite amazing.
During which time I made 60 individual bottles of media. (with help of course)
At the end of it, like a champion I go tell my boss that I had finished everything and was ahead of the proposed time, only to realise in his room that I had done only a general 20ul reaction and not the 100ul for sequencing.
He must have seen my jaw drop and eyes widen in horror at my stupid mistake because he said it was his fault for not reminding me, but we both knew it was mine. I've worked 10 months and had simply in my overconfident undergraduate level work bashing mood had forgotten the basic point of the experiment.
Which means I have to run 68 samples on a gel, and do the PCRs all over again, and inoculate 60 cultures tomorrow.
Argh.
Can someone please run me over before 7 A.M tomorrow please.

So my cousin was complaining some bacteria culture is not growing. My only advice to her was to not take bath for a few weeks. Automatica varum :D

haha! all that biology flew over my head... but funny nonetheless!
if it's any consolation... 10+ yrs into my career... I have made changes to a live database... which locked out people ALL over the world for a brief 4 hrs.
